I have the same year F150. You can move from D to 3 no problem while driving. If you're going 60 in D in 6th and move it into 3rd, you'll jump from about 1800 rpm to about 3000 rpm iirc, depending on your gearing. I've used it even when not towing, eg when passing someone just to be in gear and more into the power band before I get the opening to go around. You can use it to climb or descend hills also.
If you have tow/haul enabled, the truck will pick a lower gear for you depending on slope, how much you're braking, etc. I would let it pick the gear vs putting into 3rd unless the tranny starts to "hunt" as conditions change then maybe you just want to lock it into a gear to help keep some heat out of the transmission...
you probably want to be going slower before you drop into 2nd or 1st, but I suspect the transmission electronics won't allow it to shift into the lower gears unless safe for the transmission to do so.
