If you are talking hydraulic fluid reservoir: I ensure mine is full when all slides and level up jacks are in. I made a mark on the tank when I first picked it up (mine had no indicator marks). Then I extended everything for a normal level (no pads under jacks) and made another mark. The low level mark will change depending on how much the levelup jacks extend, but the two marks gives me fluid level reference points.
If you are talking checking the level of the RV after levelup jacks and slides are out: After being setup for weeks on unstable soil I might relevel the rig using manual and a carpenter level without bringing the slides in. The only time I releveled with auto level, yes, I brought the slides in, raised the nose with manual, and hit auto level.
