The bottom inlet is to flush the black tank, the upper inlet is for the city water, and the middle one is labeled "Anti-Freeze Inlet". When I hook the hose up to the middle one nothing happens no matter where I set the valve next to the water pump.

The antifreeze connection really isn't to be used with your water hose.
It is a port where you can attach a short piece of hose that you stick into an antifreeze jug or bucket and use the pump to distribute the antifreeze into your plumbing for winterizing.
If you do happen to hook your water hose on the antifreeze port, nothing would happen, until you turn on the water pump. And then, it would simply pump water into your plumbing, not fill the fresh water tank. (providing the valve at the pump is turned to source water from the antifreeze connection and not the fresh water tank)
My fresh water fill on my 2016 Classic Super Lite (8529IKBS) is on the opposite side of the rig from the ports in your picture. it is located on the right rear corner and is gravity fill.
