Pour in hot water and let it sit. It will hold water

Anyway, I lubricate the seal about twice a year in our Dometic 310 with food-grade silicone grease. Some use Vasoline which also works.
It is just one of those items in an RV that requires regular maintenance. Also for the non-RV people visiting, be sure they are instructed on how to properly use the toilet. Saves cleaning up a mess later.

In the Service, the head had to be clean enough to eat out of. Otherwise, one scrubs the entire barracks floor with a toothbrush.

But doesn't Vaseline contain petroleum products and you suggested earlier, was OK to use?

Perhaps he's looking out for a dog that might decide to take a drink.
I believe the bowl valve seal is neoprene and could care less if it encounters petroleum based products.

Nope, it's rubber.

I use Plumber's Grease to lube the seal. Bought it in Lowes.

But which of the 10 different compositions of rubber is it?

That's strange as most "seals" are made from Synthetics. Most common are Nitrile (NBR) and Neoprene due to both chemical resistance as well as abrasion resistance.

My issue is that the seal is off centered. I took it out and reseated it a few times, tried the hot water and that worked for a bit. Now the winter and it will not hold the antifreeze. I know the manual says to not take the toilet apart but I feel if I can get to the seal holder I can center the seal ring. I included a picture.
Is there any reason why I shouldn't separate the base?

I installed a Dometic 320 in my camper last season and it would not seal good enough to hold water for more than a day. After trying everthing to greasing it, resealing it I emailed Dometic suggesting the new seal wasn't good and they sent me a new seal which has worked great. Maybe your toilet was sitting in a warehouse for too long and the seal hardened and needs to be replaced.

Crisco vegetable shortening works well for lubricating most all of the rubber or similar type gaskets you will encounter. Use a spoon to extract enough for the job so that the cook in the camper doesn't get upset that the shortening has become contaminated.
