We really hate our camp queen RV mattress. Thin, short and uncomfortable. Technically we have enough room for a regular queen mattress, but we'll lose the walk around at the foot of the bed. This isn't a big deal for us, as we generally go through the bathroom to reach the far side of the bed. I'd need to replace the plywood under the mattress to support the full length of the mattress with a piece that's six inches longer. I'd also add a second gas strut to access the under storage.This all sounds doable to me.
Other than the increased weight of the new mattress, is there anything I'm not thinking of?

Sounds doable to me. Having your feet hanging off the bed isn't fun.

Depending on the mattress you choose, it might not increase the weight.
Friends with a regular queen in their rig replaced it with a Sleep Number bed like they have at home.

Really need a whole sheet? Add a bit to the head board side or a splice in the middle?
 
We bought a used Lexington motor home and the RV queen mattress was horrible and too short. I replaced it with a nice comfy mattress bought from Camping World during a sale. It is regular queen length (added 5-6 inches to length) and with a mattress topper we sleep like a baby! the room around our bed was non-existent anyway and we crawl onto the bed from the doorway. Sleeping good is more important than being able to squeeze around the end. The old mattress was 4" thick and new one is 8" plus a topper. I did not add any extra length support but can do so in the future if needed. We did this in May of 2022 and have done 2 very long RV trips since- 6 weeks and 7.5 weeks. The big thing to remember is opening the mattress. I was wondering how this big thing was going to arrive. It was rolled up in a square box 5' tall but about 1' square. The directors cautioned to make sure to open it with the top side up so no need to flip it once it unrolled and expanded. I guessed correctly so my wife and I use it for pleasurable wrestling matches inside of trying to flip it in the small space of the bedroom. ;)
BTW, the We-Boost is a great addition for added cell signal coverage. Most people that complain didn't read the directions. If no signal is found in remote places it can't boost. Keeping the phones within 18" of the inside antenna usually gets at least 1 bar more. Mounting to the ladder at the rear wasn't an option for where I wanted the inside antenna so I used the refrigerator chase to bring inside and then routed the coax cable into a kitchen cabinet going forward so internal antenna is just behind the cab area (class C) which is also where the recliners are where we watch TV. This allows us to use a hotspot to access streaming platforms.
 
We bought a full length queen "mattress in a box" from Wayfair and added a 2" topper. After about 25-30
nights (not in a row) we think it's pretty comfy. The only problem is it's harder to make the bed. Getting the fitted bottom sheet on the head end corners is a PIA or rather a pain in the back. Still glad we did it. Our existing gas struts still hold it up when accessing the storage even tho it's a tad heavier.
 
We replaced our OEM "cardboard" mattress with a folding version. Best investment inside the rig.
